DIECA对小麦雄性育性的影响
摘要:
为探究 DIECA(sodiumdiethyldithiocarbamate,二乙基二硫代氨基甲酸钠)对小麦雄性育性的影响,以小麦温敏雄性不育系BNS366及其近等基因系郑麦366为试验材料,在正常秋季播种(2021-10-13)和晚播(2021-12-01)条件下,于2022年春季小麦雌雄蕊原基分化期至开花期采用酶联免疫法测定幼穗和花药中内源 MeJA含量,采用0(清水,CK)、0.1、1、5、10、20和30mmol·L-1DIECA水溶液对小麦进行全株喷施处理,用I2-KI法测定花粉可育率;用国内法和国际法测定自交结实率。结果表明:在正常秋季播种条件下,在花粉母细胞期后,郑麦366内源 Me-JA含量出现了刺激性增长,BNS366则出现不足现象;1mmol·L-1和5mmol·L-1DIECA 处理对正常秋季播种郑麦366的自交结实率具有显著降低效应,5mmol·L-1 DIECA 处理效应较强,其国内法自交结实率和国际自交结实率分别比对照降低了39.82%和44.68%,在不同播期下,郑麦366和 BNS366其他 DIECA处理的花粉可育率、国内法自交结实率和国际法自交结实率与对照相比差异均不显著。DIECA可能通过抑制郑麦366内源 MeJA含量的刺激性增长,影响花药发育和正常开裂,降低了郑麦366的雄性育性,内源 MeJA 可能参与了 BNS366雄性不育的发生。
in order to investigate the efeets of sodium diethyldithiocarbamate( DlECA) on wheat male fertility, the thermo-sensitive genie male sterile wheat BNS366 and its near-isogenie line Zhengmai366 were used as materials. The two materials were sown under conditons of normal sowing in autumn(2021-10-13) and late sowing(2021-12-01), respectively. In the spring of 2022, indirect enzyme-linked immunosorbent assays method was used to endogenous methyl jasmonate( MejA) content in young panicles and anthers from the differentiation stage of pistil and stamen primordium to anthesis, and the wheat was sprayed with 7 diferent coneentrations(0(water/CK)、0.1、1、5、10
20 and 30 mmol .L-1) of DlECA solution, l2-kl method was used to detect the fregueney of fertile pollen grains at anthesis stage, National and international methods were used to determine the self-seeds rate. The results showed that under the condition of normal sowing in autumn, after the pollen mother cellstage, there was stimulating growth in the content of endogenous MejA in Zhengmai366, however, there was shortage in BNS366. Under the condition of normal sowing in autumn, sprayed with l mmol.L-1 DlECA and 5 mmol.L-1 DlEcA the self-seeds rate of Zhengmai366 were deereased which meant the male fertility of Zhengmai366 was decreased, and the treatmentof 5 mmol .L-1 DlECA had stronger effeet on it ,i.e., the self-seeds rates by national and intemational methods were reduced by 39.82% and 44.68% respeclively compared with the control group, the freguency of fertile pollen grains and the self-seed rates by national and international methods of Zhengmai366 and BN$366 under other DlECA treatments were not significantly different from the control. It was probable that DlECA inhibited the stimulating growth above mentioned, that affected the anther development and dehiscenee regularly that dereased the male fertility of Zhengmai366, and the endogenous MejA might involved the occurrenee of male sterility in BNS366.
